Il background storico del Rigshospitalet risale al primo vero ospedale della Danimarca, il "Royal Frederiks Hospital", fondato per prendersi cura e curare i poveri di Copenhagen con attaccamento. L'ospedale fu inaugurato nel 1757 e si trovava a Bredgade, dove oggi si trova il Kunstindustrimuseet. Nel 1910 l'ospedale divenne parte dello stato e si trasferì nella sede attuale in Blegdamsvej. L'ospedale ha anche cambiato nome in Rigshospitalet, poiché in futuro dovrebbe essere a disposizione di tutti i cittadini del Regno. Il moderno Rigshospital, con i suoi alti edifici grigi, risale agli anni '1960 e oggi fa parte della Regione della Capitale.
